- After they are forced to live next to a fraternity house, a couple with a newborn baby do whatever they can to take them down.

A few epic parties later, however, and Mac and Kelly have had more than their fill of the frat's wild antics. Before long the gloves have come off, and a turf war erupts between the parents who only want a solid night's sleep and the Greeks who crank up the stereo until the crack of dawn. Who will win is anyone's guess, but one thing's for sure -- this neighborhood isn't big enough for the both of them..

- Mac (Seth Rogan) and Australian-born wife Kelly Radner (Rose Byrne) are a young couple with a newborn daughter, Stella. Mac and Kelly cannot even have sex as they think that Stella is watching them, and she knows what is going on. The restrictions of parenthood make it difficult for them to maintain their old lifestyle, which alienates them from their friends Jimmy Blevins (Ike Barinholtz) and his ex-wife, Paula (Carla Gallo). Paula invites Kelly for a rave party, and she really wants to get out of the house. Mac and Kelly decide to take Stella with them, but they soon figure out that Stella has to travel with a lot of equipment, which makes it impossible for them to leave the house. The house next door to them is for sale, and Mac gets excited when a gay couple comes by for an open house.
One day, the couple finds out that Delta Psi Beta, a fraternity known for their outrageous parties, has moved into an adjacent house. The fraternity's leaders, Teddy Sanders (Zac Effron) and Pete Regazolli (Dave Franco), aspire to join Delta Psi's Hall of Fame by throwing a massive end-of-the-year party. The history of the fraternity goes back to 1930, when they threw the first Toga party on campus. In 1971, the fraternity invented beer pong. In 1985, the leader invented the Boot and Rally, when he vomited in his boot from excessive drinking, but continued to drink.
Kelly knows that this is a nightmare scenario with them having a baby. Mac and Kelly go over to introduce themselves and take Stella to make the point that they have a baby. They try to be cool and nonchalant, but Kelly asks them to "keep it down".
One night, the couple ask Teddy to keep the noise down. Teddy agrees on the condition that Mac and Kelly always call him instead of calling the police. To earn Mac and Kelly's favor, Teddy invites them to join the party, which the couple agree to. They get wasted on alcohol and drugs. At the party, Kelly meets Teddy's girlfriend, Brooke Shy (Halston Sage), and Teddy shows Mac his bedroom, which includes a stash of fireworks and a breaker box that controls their power. Mac and Kelly leave only the next morning after they all believe that they have a solid understanding.
The following night, Mac is unable to get in touch with Teddy to ask him to keep it down. Mac had already called 10 times, it was 4 am and the baby was completely freaking out. Kelly convinces Mac to call the police anonymously, but Officer Watkins (Hannibal Buress) identifies them to Teddy. Watkins says that even cops have caller ID and that is how he knew that the call originated from the Radner residence. Teddy tells Watkins that the Radners partied with them just the previous night and has the photos and videos to prove it.
Betrayed, Teddy leads Delta Psi in hazing Mac and Kelly, resulting in Stella nearly eating an unused condom after the fraternity dumps their garbage on the Radners' lawn. The couple goes to the college dean, Carol Gladstone (Lisa Kudrow), and learn that the school has a three strikes policy. Burning down their old house was Delta Psi's first strike.
Mac damages the Delta Psi house by cutting down their water pipe, which floods the basement. The fraternity takes pictures of their penises & sells it to raise $ 10,000 & make more than enough money to fix the damages.
After failing to force the fraternity to move by damaging their house, Kelly manipulates Pete and Brooke into having sex and Mac gets Teddy to catch them. Teddy and Pete fight, which ends with a barbecue grill being rolled into the path of a passing car and injuring a professor, giving Delta Psi their second strike and places them on probation, effectively ending their party plans.
Determined to shut down the fraternity, Mac and Kelly enlist the help of Jimmy, who is jealous that Paula is sleeping with Delta Psi member Scoonie (Christopher Mintz-Plasse). To acquire evidence of Delta Psi's hazing, they hire a pledge nicknamed Gary "Assjuice" (Craig Roberts) to stand up to Teddy and record him threatening retaliation. When Teddy instead shows him kindness, he reveals that Mac and Kelly hired him and are trying to sabotage the fraternity. A vengeful Teddy violently pranks the Radners and Jimmy with airbags.
Mac and Kelly send Teddy a counterfeit letter from Gladstone enabling them to have parties again as Gladstone had banned Teddy from having parties after the 2nd strike. Teddy begins planning their end-of-the-year bash.
Once the party is in full swing, the Radners call Watkins to complain about the noise. Teddy discovers the random strangers sent by Mac, Kelly and Jimmy. After finding a flyer about the party. Teddy did not circulate any fliers, & in fact it was Mac & Kelly advertising the party to catch Teddy in the act and determining the letter is counterfeit, he stops the party just as Watkins arrives. Jimmy throws himself from the balcony to distract Teddy, allowing Mac and Kelly to sneak into Teddy's bedroom and restart the party using the breaker box. Teddy catches them and fights Mac, while Kelly lights one of the fireworks and shoots it at Watkins's patrol car. Teddy takes the blame for the party and convinces Pete to take the others and flee. Gladstone shuts the house down and Mac and Kelly return home, adjusting to their new lives.
Four months later, Mac is at an outdoor shopping mall when he runs into Teddy, who is working as a shirtless greeter at Abercrombie & Fitch. The two greet each other warmly and Teddy tells Mac that he is attending night classes to complete his degree. Mac takes off his shirt and jokingly acts as a greeter with Teddy.
Mac and Kelly later take pictures of Stella dressed in various costumes for a calendar. They get a call from Jimmy and Paula, who are attending a wild party and invite the couple to come, including Stella. Mac and Kelly decline, accepting their new roles as parents.
