Well known for providing many of the female voices on The Simpsons (1989).

Aside from her voice acting work, she also provided additional vocals in three of 'Weird Al' Yankovic's songs, "Ricky" (as Lucy Ricardo), "Pretty Fly For A Rabbi" (alongside another voice actress Mary Kay Bergman) and "Jerry Springer".

She is a lifelong liberal Democrat.

She is training Debra Wilson to be the voice of Daisy Duck for when MacNeille either retires or dies.
