Forensic anthropologist Dr. Temperance "Bones" Brennan and cocky F.B.I. Special Agent Seeley Booth build a team to investigate murders. Quite often, there isn't more to examine than rotten flesh or mere bones.
Bones works at the Jeffersonian Institute. There is no such place in Washington, D.C. However, the name is probably a play on the Smithsonian Institute. Shots of the outside of the actual Smithsonian are often used on the show.
Dr. Temperance Brennan:
Don't call me "Bones".
During the title sequence, when David Boreanaz is presented, the "FBI" label on his jacket is mirrored. The entire shot is mirrored from the one actually shown in an episode, though, for artistic purposes.
