In addition to Joel (Paul Rudd) mentioning that New York City is like another character in the story, in the first scene of the movie, this idea is mentioned on every poster and advertisement, on the movie's website, and in the credits.

Molly: Listen, before you ask me anymore questions from the Icebreaker Handbook, here's the 411: Upper Sweet Side NYC. Does the name ring a bell?
Joel: Yeah. Little candy shop?
Molly: Ding ding ding! Give the man a prize, he's a winner! What does he ...
When Joel and Molly are seated in the big picture window of a coffee shop, gabbing, the sign for the subway stop across the street, although partially obscured, clearly says "__ Street Subway Station" (around the 31-minute mark). A minute later, the same sign says, "Upper W. Side Subway Station". (Of course neither sign really exists along the 2/3 line, which services my neighborhood.)
