When Sinclair and his men break into the gold car and take a blanket off the gold, one of the men shouts: "Must be a BILLION there, at least!" That was 10 times as much money, and then some, as there was in the whole United States Treasury. The script probably said "million," and the actor got over-excited.
When the gang enters the baggage car and uncovers the gold, it is stacked in layers against the right wall. A heavy gold shipment like this one would unbalance the car and might even cause it to turn over. In reality, the gold would be loaded along the center line and probably over the wheels to prevent the car floor from buckling under the weight.
As the train rolls into Dodge City, the track is unballasted (no rock anchoring it.) Track laid in such a fashion would be unstable and shift out of line quickly, making it dangerous and unusable.
The two graves that were dug by Newly and the others are perfectly smooth holes, as if they were done by construction equipment, rather than the shovels the men used.
Matt Dillon supposedly took Sinclair's left hand in a gunfight in the past. He has a leather sleeve to cover it. However Sinclair is right handed so wouldn't Dillon have shot his trigger hand blowing off his right hand back then not left hand.
Some of the voices are dubbed over and sound much louder and clearer than the voices of the other actors. Two obvious examples are the conductor and the Mexican train robber.
When examining Matt, Doc says "The bullet is lodged in the spinal column, Kitty." Then the audio is "I can't do it," but Doc's lips say, "I can't operate."
When Festus takes off with the gold-laden wagon, he should have discarded to gold bars as he drove.
That would have scattered the gold and distracted the robbers, and made the wagon lighter so it could travel faster.

When "Doc Adams" is sending the telegram to Denver, he has it signed off as "J. Adams," but it should be 'G. Adams.' Listen more closely and it can be heard that Doc does say 'G. Adams." Closed captioning also shows it this way.
The robbers back a wagon up to the door of the baggage car to start unloading the gold. The gold is on the other side of the car, right next to the door on that side. The gold, which is heavy enough as it is, has had extra metal formed into the bars to make it a lot heavier. They should have moved the wagon to the other side of the tracks so they wouldn't have to carry each bar of gold those extra feet.
